To answer your questions:
1- You can Captain Abilities when you get fouled. You get fouled when enemies Slide Tackle, or Hit you when you're -not- in possession of the ball. It's random as to which ability you'll get... you may get a Green Shell, or Bob-Omb, however, the lower your score is, the more likely it is you'll gain a Captain Ability, or another high end item.
2- Only the Captain uses Captain Abilities. If you're in control of another teammate, and activate the Captain Ability, then they'll use their Ability where they stand.
3- Captains are the only teammates who can use Megastrikes. Instead of Skillshots, they use Megastrikes, and depending on the Captain, it can be relatively easy (Daisy) or relatively difficult (Bowser) to pull them off. Good human players will block even the hottest Megastrikes, making it a true desperation move, not something to be relied on.
Dude, there's so many tips for beginners, as this game is just... deeper than it looks!
