Hey can u unlock everyone in one round of subspace emissary? i mean every single character in one play of subspace

No, once you see the character in SE, you get him/her. You don't have to fight him/her if you unlock with SE

So, yes you can unlock them all in one go, that is, if every character is in Subspace Emissary
