When you get the game (this goes for any online enabled game), you will log into the Nintendo WFC (as Nintendo calls it, Nintendo WiFi Connection), and from there you will be assigned a Friend Code. You just need to trade this code with other people to add them to your Friends List within the game. Note that you don't need to add anyone to play online, you can play against strangers on-the-fly, but there is no way to add anyone you meet in-game to your friends list.
There is also a Friend Code for the console itself. This one is used for the Message Board, so you can send messages (and screenshots from games that support it) to people in your Address Book. You can also send to and receive from regular email addresses from here as well. This code can be found in the message board (I can't recall where exactly it is, one of the Wii manuals should tell you).
There used to be a Mario Kart Friend Code trading thread here (there usually is a thread for each online game), but I think it got lost when the site crashed last week. Another one will probably pop up sometime, or you could start one yourself when you get the game. There was also a separate site built by a forum user for trading codes for all online games, but I can't be certain it is still going to be upheld since the crash (I didn't use the site myself, but perhaps someone else who has/does can chime in about it).
